Effect of Minor Antigen Disparity upon the Cellular Immune Reaction in Heart-lung Transplanted Mice




Abstract
Improved result of organ transplantation has come from the innumerable studies about the immunologic reactions occuring after exposure to transplantation antigens such as major and minor antigen, of which minor antigen has not been well
documented.
The importance of minor antigen is literally expressed by the rejection response after the renal transplantation in th HLA identical siblings.
In order to clarify the role of minor antigen in organ transplantation we performed the heart-lung transplantation in the group of both major and minor antigen disparity like BALB/C)H-2D)-CBA(H-2K), and DBA(H-2D)-CBA where the graft survivals
were
8.5
days and 13.0 days each. In the group of minor antigen disparity alone, the graft survival was 52.5 days and 52.7 days after reciprocal transplantation between C3H(H-2K) and CBA mouse.
Splenic hypertrophy was noticed from the 6th post-operative day regardless o combinations of strains.
The yield of splenochyte showed the continuous elevation from the 4th day up to the 14th day in the group of BALB-C-CBA and DBA-CBA. However the yield in the group of C3H-CBA, showed elevation from the 8th day and decreased after 11th day.
The pattern of splenocyte cytotoxicity in the group of BALB/C-CBA and DBA-CBA showed high level even in unsensitized host and elevated host, representing the inconsistent rinding with the yield of splenocyte.
In the group of minor antigen disparity alone, the splenocyte cytotoxicity positively correlating with the yield of splenocyte showed the low level in unsensitized host and elevated from the 6th to 11th day and then decreased.
The cytotoxicity of NK cell showed the similar finding with the cytotoxicity of Tc cell illustrating the sympathetic role of NK cell in early cellular immune reaction in organ trasplantation.
